Engineering Lactococcus lactis as a Cell Factory for the Production of Limonene

Limonene is a plant monoterpene which contributes significantly to the scent of most essential oils due its pleasant fragrance. The compound had been reported have anti-cancer properties against several types cancer including colorectal cancer. However, production this in nature limited because it produced as secondary metabolite. To overcome these challenges, Lactococcus lactis was developed heterologous host for limonene. A synthesized limonene synthase (LS) from Mentha spicata (mint) cloned into L. NZ9000. Western blot analysis using mouse IgG His-Tag monoclonal antibody showed successful LS expression by at size ~55 kDa. GC-MS results that optimum after 24 h induction (~8.0 ppm). Metabolic engineering attempted enhance overexpression lactococcal 3-hydroxy-3-methylglutaryl coenzyme reductase (HMGR) and mevalonate kinase (mvk) genes bacterial host. recombinant carrying pNZ:LSMM plasmid successfully enhanced two-fold (~15.1 ppm) induction. outcomes study show potential produce proteins bioactive compounds production, prospectively leads an oral delivery system compounds.
